چکیده

A rapid, high-pressure liquid-solid chromatographic technique was developed for the separation, detection and analysis of ecdysteroids. The advantages in using this system for the identification of ecdysteroids in combination with NMR and mass spectrometry and its potential application in analyzing material from biological and synthetic, systems are presented. INTRODUCTION Although the molting hormones (ecdysones) are present in small quantities in insects, riderable body of knowledge concerning their structure, metabolism, and I. 'abolic precursors has been accumulated (1, 2, 3, 4, 5). The micrk ~termination of these hormones has been carried out primarily throug_+ the use of a variety of biological assays (4). However, studies concerned with determinations such as the mode of action, specific metabolism, and excretion require that these hormones and their metabolites be isolated, separated, end identified, Such identifications have been accomplished by a combination of column ~hrorna~o~aphy~ thin-layer chromato~ap~ * NRC-ARS Postdoctoral Associate. Ecdysones and synthetic analogs: Molting hormone activity and inhibitive effects on insect growth, metamorphosis and reproduction.

Five ecdysones and 19 ecdysone analogs were assessed for molting hormone activity in the house fly assay. When these 5B-steroids were further tested in the house fly, the confused flour beetle, the yellow fever mosquito, and the German cockroach, many of the compounds inhibited growth, development and/or reproduction. The relationship of structure to both the molting hormone and inhibitive acti...

Identification of the molting hormone of the sweet potato (Bemisia tabaci) and greenhouse (Trialeurodes vaporariorum) whitefly.

In order to identify the whitefly molting hormone, whole body extracts of mature 4th instar and newly formed pharate adult Bemisia tabaci (Biotype B) and Trialeurodes vaporariorum were prepared and subjected to reverse phase high performance liquid chromatography (RPHPLC). Ecdysteroid content of fractions was determined by enzymeimmunoassay (EIA). The only detectable ecdysteroids that were pres...

The molting hormones from the embryonated EGG of the tobacco hornworm, Manduca sexta (l.).

26-Hydroxyecdysone is the predominant molting hormone in 24- to 44-hour-old embryonated tobacco hornworm eggs, accounting for approximately 80% of the ecdysones present at this stage of development. This molting hormone was previously shown to be the major ecdysone present in 48- to 64-hour-old embryonated eggs of this insect. During both of these periods of embryonic development in the hornwor...

Thin-layer chromatographic in situ analysis of insect ecdysones via fluorescence-quenching.

The possibility of quantitating insect ecdysones in situ on thin-layer chromatographic plates was examined. Two approaches were evaluated: 1) the induction of ecdysone fluorescence by sulfuric acid treatments and 2) the fluorescence-quenching of fluorescent thin-layer chromatographic plates by ecdysones. The fluorescence-quenching method was found to be most suitable and had a linear response r...

LC/MS/MS identification of 20-hydroxyecdysone in a scorpion (Liocheles australasiae) and its binding affinity to in vitro-translated molting hormone receptors.

Recent advances in mass spectrometry (MS) technology have facilitated the detection and quantification of minor components in organisms and the environment. In this study, we successfully identified 20-hydroxyecdysone (20E) in first instar nymphs (7 days after hatching) of the scorpion Liocheles australasiae, using tandem mass spectrometry combined with high-performance liquid chromatography (L...
